CHROMATOGRAPHY

INTRODUCTION:

Chromatography is a technique for dividing the components of a mixture. The stationary phase is the next substance it passes through after the mobile phase, which is the first substance.

The components of the mixture move through the stationary phase at varying rates, which causes them to separate from one another. The properties of the distinct mobile and stationary phases govern which substances move faster or slower and how they are divided. These varying travel periods are referred to as retention times.

PRINCIPLE

Based on their distinct interactions with a stationary phase and a mobile phase, components of a mixture can be separated and analyzed using the principle of chromatography. It is based on the differential partitioning principle, which separates the components of the sample by distributing them between the stationary and mobile phases.

PROCEDURE

Before we begin, let's go over the various stages of the Column Chromatography Experiment.

The mobile phase, which is composed of solvents, carries out the following tasks: acting as a solvent to enable the addition of a sample mixture to the column; acting as a developing agent to aid in the separation of components in the sample into bands; and acting as an eluting agent to remove the bands formed during the experiment from the column.

 Examples of mobile phase solvents include ethanol, acetone, water, acetic acid, pyridine, and other polar solvents.

The stationary phase is a solid material that has to satisfy the following requirements and have high adsorption properties:

Particle size and shape: Particles should be uniform in size and shape and have a diameter of between 60 and 200 microns.

Excellent mechanical stability and chemical inertness characterise particle stability and inertness. • It need to be inexpensive, accessible, and colourless.

It shouldn't react with any of the experiment's other solvents, acids, or bases.

The mobile phase should have unrestricted movement.

It ought to be able to separate mixtures of various compounds.

Types:

There are various chromatography procedures, such as:

1. Thin Layer Chromatography (TLC): This technique involves coating a solid support, usually a glass plate or plastic sheet, with a thin layer of stationary phase.

2. Gas chromatography (GC): A liquid or solid coated on an inert solid substrate serves as the stationary phase, while a gas serves as the mobile phase.

3. Liquid chromatography (LC): In this method, a liquid is maintained in a column as the stationary phase, and another liquid or combination of liquids serves as the mobile phase.

4. High-Performance Liquid Chromatography (HPLC): This technique is comparable to liquid chromatography (LC), but it uses high-pressure pumps to achieve quicker and more effective separations.

5. Ion Exchange Chromatography: Ion exchange resins are used to separate components based on how their charges interact with the resins.

APPLICATIONS

1. Chemical Analysis: Chromatography is widely used to analyse chemical compounds both qualitatively and quantitatively. It aids in the identification and measurement of the constituent parts of complicated mixes such forensic samples, natural products, food samples, environmental contaminants, and drug mixtures.

2. Pharmaceuticals: Drug development and quality control both heavily rely on chromatography. It is utilized for formulation analysis, active pharmaceutical ingredient (API) characterization, impurity determination, and drug purity testing.

3. Environmental Monitoring: Chromatographic techniques are used to track and examine pollutants in the environment, including pesticides, herbicides, industrial toxins, and volatile organic compounds (VOCs).

4-Food and Beverage Industry: Chromatography is used in food analysis to identify and measure food additives, pesticides, mycotoxins, vitamins, flavours, and fragrance components. It assures compliance with regulations, quality control, and food safety.

5- Forensic Science: In forensic laboratories, chromatography techniques are used to examine drugs of abuse, poisonous compounds, gunshot residue, accelerants in arson investigations, and trace evidence. It is beneficial in both criminal investigations and court cases.
